Outline of Annual Research Achievements

ミトコンドリア分裂促進GTP結合タンパク質Drp1と細胞骨格制御Gタンパク質tubulinに着目し、Gタンパク質の超硫黄化状態を質量分析で解析するとともに、環境親電子物質の曝露で超硫黄鎖を分裂させた際にタンパク質の機能がどう変化するか解析した。細胞に過剰発現させたflag-Drp1タンパク質を精製しSH基の超硫黄化状態をアルキル化試薬を用いて分析した結果、60%以上のCysSH基が正常状態において超硫黄化されていることがわかった。低酸素ストレスやタバコ副流煙などの環境親電子物質を曝露させることで、C末端のCys644の超硫黄鎖が高感度で脱硫黄化され、それに伴ってCys644のかさ高さが減ることでDrp1活性が増加することを計算科学的かつ生化学的に実証することに成功した(論文投稿中)。一方で、正常状態から超硫黄化されていないCys505に関しては、高濃度の硫黄ストレス曝露の際に生じるDrp1の多量性形成に関与することが明らかになった。
超硫黄分子に対して高選択的な蛍光指示薬やタンパク質超硫黄鎖検出アルキル化試薬を用いた解析の結果、心筋組織や心筋細胞内には超硫黄分子が豊富に存在することがわかった。特に虚血(低酸素)ストレス曝露において超硫黄分子の分解と、それに伴う硫黄代謝物(H2S/HS-)の蓄積が生じ、その結果、還元ストレスが誘導されること、虚血心筋障害の長期予後に対しては、グルタチオンなどの還元剤よりむしろ、酸化型グルタチオン(GSSG)のほうが心筋保護に有効であることがマウスで示された。その主たる機序として、GSSGはDrp1のCys644のグルタチオン化を促進し、虚血時に起こるミトコンドリア過剰分裂を防ぐことを示した。以上の結果は、虚血心疾患に対する治療概念を大きく覆すとともに、GSH製剤合成の副産物と考えられていたGSSGの新たな効能を示唆する画期的な知見である。

Reason

超硫黄分子の心筋細胞・組織内イメージングおよびタンパク質超硫黄化検出技術を構築し、心筋の超硫黄代謝異常が心臓の頑健性(ストレス抵抗性)低下と高い相関関係にあることを見出した。現在その成果を論文執筆中であり、2023年度中には公表できると期待できる。

Strategy for Future Research Activity

イメージング計測結果の妥当性を高めるため、質量分析技術グループと連携し、できるだけ複数の評価系で実証する。得られた成果を迅速に論文化し、超硫黄分子の概念を世界に広く発信する。Drp1多量体形成の意義については、Cys505変異マウスの作出もできており、繁殖でき次第、心不全モデルとの掛け合わせによりその意義を実証していく予定である。
