Outline of Annual Research Achievements

本研究では、人為的な機能制御と分子多様性創出の格好のモデルともいえる、糸状菌由来メロテルペノイド生合成遺伝子クラスターをとりあげた。テルペノイドとポリケタイドのハイブリッド型化合物であるメロテルペノイドは、テルペノイドとポリケタイドそれぞれの多様性を相乗した分子構造多様性を有する可能性を秘めている。有機化学を基盤として、分子生物学や構造生物学、タンパク工学の技術などを駆使することにより、メロテルペノイド生合成マシナリーを酵素遺伝子レベルで解明し、複雑多様な分子構造の構築原理を明らかにすることを目的とした。
最終年度では、Aspergillus variecolorが産生するアンジトミンの生合成マシナリーの全容を解明し、アメリカ化学会誌に論文を公表した。単離・構造決定より三十余年を経て初めて、その生合成分子基盤を明らかにするとともに、自然界の巧みな合成戦略の一端を見出すことに成功した。当初生合成への関与が予想されたDiels-Alder反応は、実際にはアンジトミン生合成に関与しないことが判明したが、一方でジオキシゲナーゼAndAが特異な分子内架橋構造の形成を担うことを解明した。さらに、生合成の最終段階においても、異なるジオキシゲナーゼAndFが酸化的転位反応を触媒しアンジトミンを与えることを見出した。現在は、これら特異な反応を触媒する一連のジオキシゲナーゼの機能をより詳細に明らかにすべく、X線結晶構造解析が進行中である。一方、Aspergillus terreusが産生するテレトニンの後期生合成段階の解明にも成功し、同じくアメリカ化学会誌に論文を公表した。
