Research Abstract

H23年度は炎症と発がんの免疫応答を解析する目的で1.起炎性疾患モデル、と2.炎症の発がん・抗がんスパイラルへの干渉査定系、の構築を計画した。
1.起炎性疾患モデルの構築
HCV感染マウスモデルの作製のために、1.PolyI:C刺激でLoxP-HCVゲノム(10kb以下)が複製開始する系(LoxPをCre recombinaseで飛ばして成熟マウスに発現できるためHCV toleranceにならない)は小原研(都臨床機構)より恵与頂き、飼育中である。2.J6-JFH1 HCV株の易感染マウス肝細胞株の樹立に成功した。IPS-1-/-マウス肝細胞にヒトCD81を発現させるとJ6-JFH1株の感染が成立した(論文投稿中)。この肝細胞株の感染効率を上げるため、非感染細胞は増殖せず感染細胞のみが増殖する系を作製中である。まずin vitroの細胞感染系を使って、HCV株のヒト、マウス肝細胞へのtropismを規定する因子群を同定する。
2.炎症の発がん・抗がんスパイラルへの干渉査定系の構築
炎症発がんモデルとして、PI3K p60tgなどの発がんマウス系を取り入れ、微生物のPAMP、内因性の起因物質(DAMP)の発がん誘導系も作製を推進中である。これらのマウス系をMyD88-/-,TICAM-1-/-,IPS-1-/-,などのマウスと交配し,がん進展、抗がん環境への自然免疫の経路の影響を査定する。移植がんの系はC57BL/6に3LL,B16D8,B16F10,EL4(EG7),などを使用して腫瘍浸潤マクロファージの寄与を解析している(論文投稿中)。樹状細胞,genechip,proteome,やyeast two-hybridなど抗がん因子を同定する方法を立ち上げた。
