Outline of Annual Research Achievements

(1) sPLA2-VとsPLA2-IIEは肥満の脂肪細胞に発現誘導され、全身の代謝状態を制御するMetabolic sPLA2であること、またsPLA2-VがTh2応答を誘導する唯一のTh2-prone sPLA2であることを論文発表した（Cell Metab 2014）。(2) sPLA2-III（Anaphylactic sPLA2）の新しい機能として、sPLA2-IIIの欠損により肥満や動脈硬化が改善する、大腸癌が改善する、接触性皮膚炎やアトピー性皮膚炎が増悪する等の表現型を見出した。(3) sPLA2-IID（Resolving sPLA2）の免疫抑制作用の普遍性を検証した結果、sPLA2-IID欠損マウスでは接触性皮膚炎（Th1応答）に加えて乾癬（Th17応答）や肥満（脂肪組織の慢性炎症）等が増悪すること、逆に過剰発現マウスでは抑制されることを確かめた。また、欠損マウスでは腫瘍免疫が賦活化されるため、皮膚癌が抑制されることを発見した。(4) sPLA2-Xがω３脂肪酸由来の抗炎症性脂質メディエーターを動員して潰瘍性大腸炎の保護に関わることを見出した。(5) sPLA2-IIF（Epidermal sPLA2）の欠損が乾癬や皮膚癌などの表皮肥厚疾患を改善すること、本酵素の責任脂質代謝産物がplasmalogen-LPEであることを発見し、現在投稿中である。(6) 新規iPLA2であるPNPLA7はリゾホスファチジルコリンをグリセロホスホコリンに代謝するリゾホスホリパーゼであり、欠損マウスは下流のコリン・メチオニン代謝が乱れるため、脂肪肝・脂肪萎縮・早期老化の表現型を示すことを明らかにした。(7) 皮膚特異的な新規iPLA2であるPNPLA1の欠損マウスが魚鱗癬を発症することを見出し、本酵素が皮膚セラミド代謝を制御することを見出した。
