Outline of Annual Research Achievements

NF-kBのRelBサブユニットは樹状細胞の成熟に重要な役割を果たすが、その分子機構を解明するため蛍光タンパク質と融合したRelB-VenusをrelB遺伝子座から発現するノックインマウスを作成した。RelB-Venusマウスには異常は観察されないことからこのマウスを用いて一細胞レベルでのRelBの発現量と活性化と樹状細胞分化との関係を解析した。その結果、樹状細胞にはRelB-, RelBlow 及びRelBhigh分画が存在することが明らかとなった。RelBhigh分画はRelB核移行により活性化され樹状突起を出していた。一方でRelBlowとRelB-の樹状細胞はともに樹状突起がないもののMHCIIや共刺激分子の発現はRelBlowの方がRelB-より高かった。即ち、我々は、樹状細胞の中間的な成熟段階であるRelBlow分画の同定に成功した。また、ヒト白血病ウイルス１型(HTLV-1)による白血病発症に重要な役割を果たすTaxタンパク質によるNF-kB活性化機構を解析し、Taxタンパク質によって誘導される混合型ユビキチン鎖がNF-kBの恒常的活性化を誘導することを見出した。さらにin vitroのユビキチン合成実験からTax自身がE3酵素として働く可能性を検討したが、Ubc13存在下でもTaxによりK63型ユビキチン鎖は合成されず他のE3酵素の関与が強く示唆された。
