Outline of Annual Research Achievements

本研究の目的は、次世代のフィジオーム・システムバイオロジーシミュレーション基盤を確立することにある。また理論研究と実証研究を有機的に実施し、心臓の電気活動の分子レベルから組織・臓器レベルまでの統合的な理解を得ることも目的としている。
昨年度に引き続き本年度も、生理機能の多階層的モデル構築をサポートするためのプラットフォームであるPhysioDesigner、ならびにそれらのモデルシミュレーションを実行するアプリケーションFlintの研究開発を進めた。平成24年4月に第1版のアップデート版である1.0betaをリリースした後、約半年のサイクルでマイナーアップデートを繰り返し、平成25年1月に1.0beta3版を公開した。既に100名を超える研究者によってダウンロードされている。さらにプラットフォームソフトウェアの普及活動に努めた結果、physiodesigner.orgは国内外の研究者から継続的にアクセスを受けている（150view/週）。
一方、心臓の拍動リズム制御に関わるG蛋白質制御カリウムチャネルやhERGチャネルの機能を実験科学的に解析すると共に、数理モデル化とシミュレーションによる理論研究を進めた。in vivoの心筋細胞ではリガンドへの感受性や不活性機構に差のある２グループのACh受容体およびG蛋白質制御カリウムチャネルの存在が、シミュレーション研究から示唆された。また昨年度詳細に解析した薬物のhERGチャネル開口促進作用の数理モデル解析から、この作用は薬物の催不整脈作用を抑制する可能性があることが分かった。さらに遺伝子変異や薬物の未知の効果を酵母の生育という指標で、研究室レベルで簡便に効率的に予測するための新規評価系の開発にも発展的に取り組み、不整脈関連遺伝子異常や薬物効果を検出できるイオンチャネル機能評価系を立ち上げることが出来た。

Reason

統合環境プラットフォームソフトウェアPhysioDesigner/Flintの完成度を高めることができ、改訂版を一般に公開した。これにより、領域内の研究者のみならず、領域外の研究者も最新のPhysioDesigner/Flintの利用が可能となった。ユーザーからのフィードバックも得られ、開発速度はより高まっている。順調に進展していると言える。
また、心臓電気現象の統合研究においても着実な進展が見られる。心臓の拍動リズム制御や興奮伝導に重要な役割を果たすイオンチャネルの機能解析が進み、原理に基づいた拍動制御や不整脈リスクの評価がin silicoで可能になった。さらに、酵母を用いたイオンチャネル機能評価システムを開発できたことから、今後はイオンチャネルの機能異常をもたらす遺伝子異常や薬物を従来よりも簡便に検出することが出来るようになり、他の知見と組み合わせることにより不整脈のリスクについて効率的に予測することが可能になりつつある。

Strategy for Future Research Activity

これまでの研究は計画通り順調に進行しており、特記する問題点は生じていない。今後も研究計画に沿って研究を実施することで目的を達成できると考えている。
今後、PhysioDesigner/Flintの研究開発は、医用画像データも含めた形態学的データを数値計算可能なモデルに統合する機能、シミュレーション結果を可視化するための機能等を拡充する次のフェーズに入る。さらに、 PhysioDesigner/Flint利用の促進を図るために、操作性の更なる向上、モデルデータベースの拡充、チュートリアルの積極的な実施を行う。
一方、当初の計画にはなかったが、平成24年度に新たに立ち上げた酵母を用いたカリウムチャネル機能評価系から新たな知見が得られつつある。解析に必要な手順をより減らし、チャネル機能異常の検出感度を高めるように実験系を改良する。その他、従来から実施されているG蛋白質制御カリウムイオンチャネル・遅延整流性カリウムイオンチャネルの電気生理学、薬理学的な解析を引き続き実施し、シミュレーションによる解析も行うことで不整脈の発症や薬物治療に関する理解を深める。
