| Outline of Annual Research Achievements |
イオンチャネル型グルタミン酸受容体(iGluRs)は中枢神経系における興奮性の情報伝達を担う、主要な受容体の一つである。記憶、学習などの高次脳機能や視覚の情報伝達に関与する一方で、脳虚血疾患における神経細胞死、アルツハイマー病やパーキンソン氏病と密接に関連していることが最近の研究において示唆されている。
iGluRサブタイプに選択的に作用するリガンドを提供することができれば、サブタイプの役割を調べるための有用な研究ツールとなりうる。本研究では、天然由来のiGluRリガンドであるアクロメリン酸Ａとカイトセファリンをテンプレートとする、iGluR選択的リガンドの創製を目的とした。
最終年度にあたる本年度は、アナログ合成、iGluRサブタイプ結合親和性の評価、構造活性相関研究、受容体・リガンド結合様式の解析を含む、包括的な研究を実施した。特筆すべき成果として、7-epi-カイトセファリンが天然物を凌駕する優れたNMDA受容体選択的なリガンドであることを見出した。これにより当初掲げた研究目標を達成できた。
