| Outline of Annual Research Achievements |
（１）阻害剤の化学合成：テトロドトキシン（TTX）アナログの化学合成では、本年度、8-epi-TTXの合成を検討した。その結果、これまで当研究グループで開発してきたTTX の合成方法論では、合成困難であることがわかった。このアナログの合成には、別の合成法論の開発が必要である。
サキシトキシン（STX）アナログの化学合成では、昨年度までにカスケード型ブロモ環化反応によってSTXのspiroアミナール構造を合成し、その後、環状ウレアを含む三環性中間体への変換に成功した。残る課題は、環状ウレアのグアニジンへの変換だけであった。ウレアをBn基で保護された中間体では、この変換が困難だったが、この保護基をPMBとしたものでは、グアニジンへ変換され、STXの天然アナログの一つdecabamoyl-STX-olの合成を達成した。
TTXとの構造類似性から、海産天然物クランベシンBのカルボン酸部分の合成を検討し、カスケード型ブロモ環化反応を鍵反応とするルートでの合成に成功した。
（３）ナトリウムチャネル（Nav）の阻害活性評価：昨年度から検討してきた手動によるパッチクランプ実験を立ち上げることに成功し、これまでにHek293T培養細胞にそれぞれNav 1.1, 1.3, 1.4, 1.5, 1.7 を発現させ、その阻害活性の測定が可能になった。これを使って TTXのアナログの阻害活性を測定したが、5-deoxyTTX以外のアナログは、どのサブタイプのNavも阻害しなかった。また、高いサブタイプ選択性を示すアナログも見い出されていない。一方で、クランベシンBのカルボン酸は、Neuro-2A細胞を使ったアッセイ系で、 TTX に匹敵する強力なNavの阻害活性を示す事を初めて明らかにした。
