Research Abstract

本研究では,環境制御型電子顕微鏡内でのナノスケールロボット操作技術を発展させ,機能性を有した細胞を高効率に評価し,マイクロチップ内での並列型細胞播種により3次元細胞システムを超高速でアセンブリする事を目的としている.
具体的には,(1)細胞内外のインピーダンス特性,pHや温度の物理・化学量を高速・高効率に計測・制御するナノデバイスを構築する.(2)細胞・足場複合体からなる細胞システムに対して,ナノデバイスを活用し細胞及び足場間のインターラクション(摩擦抵抗力・機械的強度・付着力)を評価する.(3)超高速で細胞を3次元形状にアセンブリするため,並列型細胞播種システムを構築する,ための手法及びシステムを研究開発している.
本年度は,(1)誘電泳動力を用いた複数細胞のパターニグおよび操作技術,(2)細胞包埋型ハイドロゲルマイクロ構造体の作製手法,(3)血管構造を模擬した血小板産生用マイクロチップの開発,などについて研究開発を実施した.

2: Research has progressed on the whole more than it was originally planned.

Reason

マイクロ・ナノメカトロニクス技術を用いて,単一細胞,細胞間ネットワーク,細胞組織を計測・制御するためのマイクロ・ナノデバイスを創製した.主な成果を次の通り,記述する.
1)マイクロチップ中に組み込んだマイクロ電極を用いて,誘電泳動力を用いた複数細胞のパターニグおよび操作技術を実験的に検証した.また,紫外線硬化性材料材料を任意形状に成形する技術と併用することにより,細胞包埋型マイクロ構造体を作製する手法について,実験的な検証した.
2)マイクロ流体チップを用いて,細胞包埋型ハイドロゲルマイクロ構造体を作製する手法を実験的に検証した.また,作成したハイドロゲル構造体を,生分解性の3次元細胞足場上に固定することにより,細胞を足場上に播種する手法について,実験的な検証をした.
3)巨核球を固定するマイクロ流体チップを製作し,巨核球の固定と血小板創成を確認した.循環培養システムを構築し,連続的な血小板産生のための実験環境を構築した.

Strategy for Future Research Activity

今後の主な研究課題について,下記の通り列挙する.
1)細胞包埋型マイクロ構造体の3次元アセンブリ手法を提案する.例えば,マイクロ流体チップ中での自己組織化を利用することにより,高速かつ高効率な複数のマイクロ構造体のアセンブリを実現する.
2)細胞包埋型ハイドロゲル構造体を,高速かつ高効率に3次元細胞足場上に固定するためのシステム構築を行う.
3)動物細胞を培養・アセンブリするための実験手法を確立し,実験室内でコンタミネーションなどの問題を発生せずに3次元細胞構造体を作製する.
