Outline of Annual Research Achievements

単一細胞の局所計測・制御用のナノデバイス技術を応用し,環境制御型電子顕微鏡及びマイクロチップ内でのナノロボット操作技術を発展させ,3次元細胞システムの機能解明のためのナノスケールの高速・高効率細胞応答計測・操作システムを構築する.
(1)細胞内外のインピーダンス特性,pHや温度などの物理量により細胞の活性を高速・高効率に計測・制御するためのナノデバイスを作製し,細胞の特性や状態を計測・制御する.(2)ナノデバイスを活用することにより,細胞及び足場間のインターラクションを計測し,細胞・足場複合体のIn vivo応用のためにIn vitroにおける評価技術を確立する.(3)3次元機能性複合組織を形成するために,細胞・足場複合体のIn vitro培養及び摩擦抵抗力・機械的強度・多層間付着力などの計測・評価技術を確立する.
そこで今年度は,前年度までの成果に基づき,主に以下に挙げる研究課題について実施した.
(1)3次元細胞構造体の組み立てをスケールアップし,より簡便な操作で実現することが望まれている.そこで,磁性微粒子を細胞マイクロ構造体中に包埋し,着磁させることで磁化し,自己組織的に組み立てる手法を提案し,基礎的な知見を得た.(2)ゲル中に包埋された細胞は,培地交換を行わなければ,細胞死に至る可能性がある.そこで,アルギン酸ファイバーを溶かし出すなどの新たな手法を提案し,細胞を包埋したゲル中に毛細血管状のマイクロチャネルを作り出すための基礎的な知見を得た.(3)バイオプリンティング技術では連続的に細胞を打ち出すことが出来るが,単純に基板に打ち出したのみでは,表面張力などの影響により,細胞のパターニングが困難となる.そこで,生体適合性・生分解性材料状にマイクロパターニングを施し,バイオプリンティング技術を応用して細胞のパターニングをする手法を提案し,基礎的な知見を得た.
