Outline of Annual Research Achievements

これまでの名古屋大学医工連携、岐阜薬科大学との共同研究によるプラズマがん治療の研究の成果及びプラズマ活性溶液の成果をまとめ、レビュー論文として国際雑誌に数報公表した。具体的には産婦人科との共同研究により超高電子密度大気圧プラズマが卵巣癌治療に有効であることを示したことを紹介し、先端医療臨床研究支援センターとの共同研究により、プラズマを照射した培養液をプラズマ活性培養液と名付け、プラズマ活性培養液がアストロサイト正常細胞に対して、グリオブラストーマ脳腫瘍培養細胞を選択的に殺傷すること、プラズマ活性培養液が抗癌剤耐性の卵巣癌に対してin vitro及びin vivoで抗腫瘍効果を示すことなどプラズマ活性培養液のがん治療への有効性を紹介した。更には産婦人科及び消化器外科との共同研究によりプラズマ活性培養液が卵巣癌、胃癌、膵癌などの腹膜播種治療への臨床応用に向けた取り組みを紹介した。また眼科との共同研究により、プラズマ活性溶液は、がん治療のみならず、加齢黄斑変性の治療にも有効であることを示したことを紹介した。また、プラズマやプラズマ活性溶液が抗腫瘍効果をもたらす作用機序について、これまでに我々のグループ及び世界の研究者が明らかにしてきたことを平易にまとめた。脳腫瘍培養細胞においてはプラズマ活性培養液が生存・増殖シグナリングネットワークを抑えることにより、アポトーシスを誘導することの発見、A549肺癌細胞に対してプラズマ活性培養液がカスパーゼ非依存性のシグナル伝達経路を作動させていることの発見の紹介や、生体反応病理学との共同研究によりプラズマと細胞・組織との相互作用として、脂質過酸化等の酸化ストレスの影響を報告したことの紹介等を行った。
また、名古屋拠点プラットフォームでは総括班との連携で、ワーキンググループで進める各種プラズマソースの活性種分析を進め、分析データを各班に提供した。

Reason

これまでにA01堀班で開発されたプラズマ活性溶液中の何が抗腫瘍効果に影響を及ぼすのかという疑問はプラズマ医療における最も重要な研究課題の１つである。プラズマ照射中の反応ダイナミクスの解析に加え、最終安定化学種の生成を定量評価した結果、過酸化水素と亜硝酸イオンの比率がプラズマがん治療において特異的に効果をもたらす知見を得た。活性窒素種の気相からの供給が、硝酸比率が低い亜硝酸を形成するもの特徴が見いだされた。正常細胞に対する抗腫瘍効果の選択性が、過酸化水素と亜硝酸の相乗効果による新たなメカニズム解明が進んだ。
また、前記超高電子密度大気圧プラズマ活用のプラズマ活性溶液を使用し、A03吉川班と共同でプラズマがん治療の研究を精力的に進めてきた。その過程で開発されたプラズマ活性溶液（Plasma-activated medium, PAM）の臨床応用に向けて、名古屋大学医学部、岐阜薬科大学との共同研究による連携が深まり、多くのオリジナル論文を公表してきたが、それらの成果をまとめ、レビュー論文として国際雑誌に数報公表した。プラズマ活性溶液の成果は世界にもインパクトを与え、世界各国から招待講演を受けるようになった。更には世界中のプラズマ医療の研究者がプラズマ活性溶液のがん治療へ向けた研究やプラズマと溶液との相互作用の研究を追従するようになり、我々の論文の引用件数も増えた。プラズマがん治療研究において、世界に先駆けた成果を多く残し、レビュー論文のオファーをいくつも受け、国際会議などからの招待講演のオファーも多く受けるようになったことは、期待以上の成果と言える。
更に、プラズマから生体にいたる分子機構の解明に関してもA01堀班及びA03吉川班が新学術領域内のあらゆる班と連携することにより爆発的に進んでおり、プラズマ及びプラズマ活性溶液が細胞に及ぼす影響の統一的な理解へ向けて大きく前進した。

Strategy for Future Research Activity

ワーキンググループにまとまって各班のプラズマ源の特徴を分析し、プラズマ医療の創成に向けた医療効果の学理、作用機序の解明を進めている。プラズマ作用素について、放電させる電気的特性、放電により発生するプラズマの電子的特性、さらに化学反応を生じたラジカル組成・イオン数、電子密度が、各種プラズマ源の比較により、本質的にプラズマ医療に要求されるユニーバサルパラメータの追求に向け研究を推進する。
引き続き、国内及び国外での国際会議等における波及活動と多くの共同研究の機会を得るように努める。特に名古屋大学内では、先端医療臨床研究支援センターにより先端医療研究から産まれた新しいシーズを臨床応用へと進めてゆく体制が存在するので、新学術領域「プラズマ医療科学の創成」の最終年度で前臨床までシーズを温めてゆき、臨床応用へと橋渡しを行う。また、PAMの臨床応用に向けた次世代PAMの研究開発や規格化を進める。医療応用プラズマとしての装置開発、化学療法としてのプラズマ活性溶液の研究開発の2面から研究開発を進める。様々な条件で作成されたプラズマ活性溶液が様々な細胞に対してどのような分子に影響を与えながらアポトーシス等の生理作用を引き起こすのかの知見が蓄積されてきたので、更にプラズマ及びプラズマ活性溶液のがん細胞に対するアポトーシス誘導機構と選択的殺傷機構をより深淵かつ統括的に理解することを目指す。また殺傷と再生・増殖を制御するプラズマの条件を探索しその細胞内分子機構の解明を目指す。
最終年度であるので、これまでの成果を教科書としてまとめるために断片的な発見に関する統一的な解釈を行ってゆく。新学術領域「プラズマ医療科学の創成」のプロジェクトが立ち上がってから世界でのプラズマ医療の研究開発は目覚ましく進み我が国がこの分野を先導することに成功してきたと言えるので、より具体的にその歴史的な意義をまとめる。
