Outline of Annual Research Achievements

本研究は加齢に伴う免疫系恒常性と応答性の変容を、免疫系機能の中枢にあるT細胞系に焦点を当てて解析し、免疫老化が個体の恒常性維持と病態発生に与える影響を明らかにすることを目的とする。繰り越し経費による最終年度は、私達が発見報告したユニークなT細胞集団 (Senescence-associated T cells, SA-T)の生理的加齢にともなう増加のメカニズムの検討を進めた。若齢マウスでの胸腺摘除や老齢マウスでの胎児胸腺移植などを用いた解析の結果、加齢にともなうSA-T細胞の増加と蓄積には、生理的胸腺退縮とそれに伴うT細胞の恒常性増殖の亢進が一義的に重要な役割を果たしていることが明らかになった。また、SA-T細胞が選択的にCD153分子を構成的に発現することから、生体内でCD153を特異的に除去しうる活性を有する新たな抗CD153抗体の作成を進め、体内投与によって効率的かつ選択的にSA-T細胞を除去しうるモノクローナル抗体の確立に成功した。さらに同抗体の投与により、ループスモデルマウスにおいてSA-T細胞の増加がほぼ完全に抑制され、同時に自己抗体の産生やループス腎炎の発症がほぼ完全に予防および治癒されうることが示された。これらの結果はSA-T細胞が全身性自己免疫病の発症に重要な役割を担うことを確認するのみならず、ループス病を含む多様な加齢関連慢性炎症疾患の制御の画期的な方途の展望を開くものである。今後は、多様な前臨床試験などにより、SA-T細胞の制御による疾患制御が重要な課題になるものと考えられる。
