Outline of Annual Research Achievements

本研究は、クロマチン動態の正確な「計測」と再構成ポリヌクレオソームを用いた「再構築」を行い、生細胞や生物個体におけるクロマチンので動的変化とその機能発現における意義を明らかにすることを目的として行っている。
本年度は、ヒストンH4K20モノメチル化(H4K20me1)特異的mintbody(modification-specific intracellular antibody)を用いた解析を中心に進めた。H4K20me1-mintbodyとmCherry融合PCNA(proliferating cell nuclear antigen;DNA複製蛋白質)を発現する雌マウス細胞を樹立し、H4K20me1が濃縮する不活性X染色体の複製タイミングを解析した。その結果、条件的ヘテロクロマチンである不活性X染色体は、S期のちょうど中間で複製を開始し、その2-3時間後に複製を終了することが明らかになった。不活性X染色体の複製後には、構成的ヘテロクロマチンが約2時間かけて複製してS期が終了した。また、核と細胞質のH4K20me1-mintbodyの蛍光強度を比較することで、細胞周期に依存したH4K20me1レベルの変化を追跡することも可能となり、H4K20me1レベルはM期にピークに達し、S期の中期まで徐々に低下したのち、G2期に急激に上昇する様子が計測できた。また、H4K20m1-mintbodyの構造解析と変異体解析を行い、細胞内で機能的発現に重要なアミノ酸を同定した。これらの結果は、H4K20me1のダイナミクスの生理的意義の解明にとって重要であるほか、新たなmintbodyの開発にとっても重要な成果といえる。
ヒストンバリアントの解析においては、精巣特異的なH3バリアントであるH3tのノックアウトマウスの解析を行い、H3tが生殖細胞の分化に必須な役割を果たすことを明らかにした。

FabLEM法やmintbodyを用いた生細胞解析により、クロマチンダイナミクスと転写制御に関する多くの新規知見が得られている。

Strategy for Future Research Activity

これまでの研究体制を継続し、さらに発展させる。具体的には、培養細胞を用いた系で転写活性化や細胞分化に伴うクロマチンダイナミクスの詳細な解析を行う。また、動物個体を用いた系で発生や分化に伴うクロマチンダイナミクスを明らかにしていく。
