細胞分化にともなうクロマチン変動メカニズムの解明

Outline of Annual Research Achievements

平成27年度では、研究領域当初より予備検討を行っていたマウス及びヒトヒストンバリアントの網羅的同定を論文成果として発表を行った。概要としてマウスにおいて14種、ヒトでは3種の新たなヒストンH3バリアント遺伝子を同定した。更に転写産物の解析を行った結果、そのうちマウスについては6種、ヒトについては3種全てのmRNAレベルでの発現を確認した。タンパク質レベルにおいては少なくとも4種について確認した。更に、それぞれの細胞生物学的、生化学的な特性の解明を行った結果、精巣特異的に発現するH3バリアントH3tをはじめ、7種はクロマチンに安定的に取り込まれることからヌクレオソーム構成因子として機能しうることが示唆された。また、ヒトH3バリアントについても領域内の共同研究を展開し、H3.Y及びH3.5の解析を行い論文発表を行った(Kujirai et al. Nucleic Acids Res. 2016, Urahama et al. Epigenetics Chromatin 2016)。また、ゲノムワイドなヌクレオソーム配置及びその特性についても解析を行った。特に転写因子結合周囲のヌクレオソーム配置パターンが主に5種に大別されることを明らかにし、転写活性化状態がヌクレオソーム配置パターンより予測可能であることを示し論文発表を行っている(Maehara et al, Sci Rep. 2016)。

網羅的に同定されたヒストンバリアントの機能解明を進めていき、種特異的なヒストンバリアントの意義について理解を深めていく。以上研究計画の変更はない。
