Outline of Annual Research Achievements

多くのパターン認識受容体はマクロファージなどのミエロイド系細胞に発現し、外来病原菌由来分子を認識する一方、異常自己の認識に寄与することも報告されているが、内因性リガンドの同定は遅れている。
本研究では、特定の樹状細胞を認識する受容体を見出した。この活性は、高濃度のマンノースの添加及び受容体のリガンド結合配列への変異の導入によって消失した。次に、受容体と結合する分子を同定するため、ヒト免疫グロブリンのFc領域とを融合させたキメラタンパク質を作製し結合する分子の精製を試みた。その結果、70 kDa付近にDectin-2に特異的に結合するタンパク性分子の存在が示唆され、精製後質量分析法による解析を試みたところその候補としてβ-glucuronidaseが同定された。HEK293T細胞に強制発現させたβ-glucuronidaseでも、受容体と結合することが確認されたが、欠損マウスを作成しても活性が消失しなかったことから、機能を代替する他の候補分子の存在が示唆された。そこで、様々な系統のマウスを検討した結果、本活性が消失するマウス系統を見出した。野生型と本系統マウス由来で発現遺伝子発現を比較した結果、細胞膜タンパク質の候補遺伝子としてスカベンジャー受容体を見出した。
以上の結果より、レクチン受容体が内在性タンパク質の糖鎖の状態をモニターし、異常な自己細胞の感知に寄与している可能性が強く示唆された。

Strategy for Future Research Activity

上述の受容体以外にも、複数の活性化型C型レクチン受容体が様々な状況下で自己由来細胞を認識することを見出している。レポーター細胞、Ig融合タンパク質を用いて、リガンド発現細胞集団を特定し、生化学的、分子生物学的手法を用いてリガンド分子の探索を行い、生体が異常自己を認識する分子機構の解明を目指す。
