Outline of Annual Research Achievements

我々の研究グループは、遺伝子操作を利用して神経回路を操作・改変する新規技術の開発を進めてきた。本研究では、我々の遺伝子改変技術をさらに発展させ、第一に、神経回路の経路選択的な活動制御を目的に、光や化学物質に応答する膜タンパク質を目的の神経路に導入し、任意の時期に活動を調節する新しい技術（光遺伝学および化学遺伝学）の開発に取り組み、第二に、学習のプロセスに伴って回路が機能シフトをおこす神経機構について、大脳皮質―基底核ループを介するメカニズムの解明に取り組むことを目的にしている。本年度は、細胞特異的な光遺伝学技術を応用し、遺伝子改変ラットの線条体における直接路・間接路細胞を同定し、行動課題中の特定神経細胞の活動を記録する実験系を確立した。一方、脳固定下でのラットにおいて、刺激（聴覚）弁別学習の獲得は、困難であることが判明したため、来年度以降は自由行動下での弁別学習課題に切り替え、この課題中の直接路・間接路ニューロンの活動を記録することに取り組むこととする。細胞種特異的な化学遺伝学による経路選択的な活動制御のため、ウィルスベクターとレポーターラットを利用して、線条体の直接路・間接路細胞において昆虫イオン透過型受容体あるいは線虫イオンチャネル遺伝子を発現させる実験系を確立した。背内側線条体(DMS)の間接路に線虫イオンチャネルを発現するラットにイベルメクチン(IVM)を注入し、細胞活動を抑制することにより、刺激弁別行動の消去に変化が認められることを明らかにした。また、本年度に、PET解析によって、聴覚弁別学習の獲得過程において後部の線条体領域の活動が向上することを見出し、この領域の破壊実験により学習形成が阻害されることを明らかにした。今後、薬剤による抑制を行い学習形成への影響を探るとともに、後部線条体への入出力関係を明らかにし、選択的な経路の操作に取り組む計画である。

Reason

光遺伝学および化学遺伝学の技術を、線条体直接路、間接路の機能解析に応用するまでの技術を確立し、自由行動下での刺激弁別行動の解析のセットアップを進めた。細胞種特異的な化学遺伝学による経路選択的な活動制御のため、線条体の直接路・間接路細胞において昆虫イオン透過型受容体あるいは線虫イオンチャネル遺伝子を発現させる実験系を確立した。PETにより同定した、学習過程で脳活動が変化する部位が実際に機能的も重要な役割を持つことを明らかにすることができた。

Strategy for Future Research Activity

今年度は自由行動下での弁別学習課題に切り替え、この課題中の直接路・間接路ニューロンの活動を記録することに取り組む。Tac1-Cre、D2R-Creラットに、ウィルスベクターを注入し、組み換え反応を介して、細胞タイプ特異的にチャネルロドプシン遺伝子の発現を誘導する。レーザー刺激により活性化する神経細胞を同定し、自由行動下の聴覚弁別課題遂行中の活動を記録する。細胞種特異的な化学遺伝学による経路選択的な活動制御のために、ウィルスベクターかレポーターラットを利用して、DMS直接路およびDLS間接路・直接路に化学遺伝学による操作を施し、弁別学習への影響を解析する。また、学習プロセスの過程で起こる脳内の機能シフトの動態を把握するために、陽電子放射断層撮影法 (positron emission tomography; PET) を用いて脳活動の時間依存的な変化の解析を進め、その過程で変化する後部線条体領域の機能を損傷技術により確認したが、さらに、この領域の活動が必要な時期を定めるために、薬剤による抑制を行い学習形成への影響を探るとともに、後部線条体への入出力関係を明らかにし、選択的な経路の操作に取り組む。
