Outline of Final Research Achievements

Transcriptome studies in eukaryotes have uncovered a large number of long noncoding RNAs (lncRNAs) in their genomes. Physiological and biochemical roles of these lncRNAs are enigmatic, and thereby comprehensive understandings of lncRNA function is far away from the goal. In current study, we chose Drosophila Lobe-less (LOL) RNA as a model, which is specifically expressed in proliferating neuroblasts during embryogenesis. Using various nerve cell specific cell markers, we found that the lol gene is essential for axon guidance of mushroom body neurons. Transcriptome analysis with a high-throughput sequencer showed that lol regulates many genes including neurogenesis- and axon-guidance specific genes in. We also found lol genetically interacts with Polycomb Group (Pc-G) genes, suggesting that lol functions as chromatin modifiers, consistent with its nuclear localization.

Academic Significance and Societal Importance of the Research Achievements

Lobe-less(LOL)RNAはショウジョウバエの中枢神経系に発現する長鎖ノンコーディングRNAであり、LOL RNAを欠損した変異体では、行動や記憶を司る脳内中枢であるキノコ体の部分欠損が起こることが知られている。本研究ではlol変異体の詳細な解析を行い、lol変異体の表現型がキノコ体神経細胞の軸索誘導の異常に起因するものであること、実際にlol変異体では多数の軸索誘導関連遺伝子の発現に異常が見られることを示した。これらの結果は、LOL RNAのみならず、多数見つかっている他の神経系特異的長鎖ノンコーディングRNAもまた神経発生において重要な機能を担っていることを示唆するものである。
