我々は、脳タンパク質老化の進展メカニズムの解明と精神・神経変性疾患の治療を目的として、生体脳イメージング技術を駆使した治療評価系の開発を目指している。２年目の本年度よりタンパク質構造化学に裏付けされたPET薬剤開発を進めている。現在、生体イメージング技術を用いたマウスモデルの治療評価系の開発を進めており、この評価系を活用した創薬プラットフォームの構築を目指している。我々の研究成果として、タウPETイメージングによる生体脳でのタウ病理像が形態MRIで見られる脳萎縮と相関することが確認されており、PETイメージングで検出される神経炎症とも関連性が高いことが明らかとなった。また、我々の用いるタウPETリガンドが蛍光特性を有することから、二光子顕微鏡による細胞レベルのタウ病変進行過程を生体内で追跡することに成功した。これらのマルチモーダルイメージング技術をさらに発展することで、動物モデルによるタウオパチーに対する早期治療評価系を確立できると考えている。また、並行してマーモセットの脳タンパク質老化モデルの作出、核磁気共鳴法による老化タンパク質の構造解析、新規イメージングバイオマーカーの開発も進めている。

Reason
動物モデル、PETイメージング、タンパク質構造化学解析を順調に進めており、マーモセットモデルの作出に若干の遅れが生じているが、マーモセット脳のMRI用コイルが使用可能となったことから、脳機能計測に関わる実験が本年度より開始されており、概ね順調に進展している。
