| Research Abstract |
前年度の研究を継続、進展させ、下記の成果が得られた。
1.AFPの糖鎖が産生細胞により異なる事を明らかにし、肝硬変症、肝癌、卵黄のう癌のAFPの分別測定を可能とし、新しい診断法を開発した。多くの臨床例において検討しこの事実を確認した。肝硬変症よりの肝癌発生を早期に診断可能な事を示した。
2.AFPcDNAのイースト菌、培養動物細胞での発現産物は天然のAFPと同様の生物活性を示した。AFPのエストロゲン結合部位を特定した。各種培養細胞へAFPcDNAを容易に導入し得る系を開発したが、その発現に細胞特異性を認めた。
3.CEA、AFPに対する抗体での放射免疫局在診断を数十症例で行い、半数で腫瘍像が描出された。より適した抗体の選別、標識核種、およびフラグメント化抗体の検討を行った。
4.AFP抗体と制癌剤の複合体を作製し、in vitro、in vivoのラットおよびヒト腫瘍で制癌効果を立証した。数十例の臨床例で制癌剤とモノクロナル結腸癌抗体(A7)の複合体に制癌効果を認めた。
5.CEA遺伝子ファミリーの産物を識別するモノクロナル抗体を作製した。CEAおよびNCAの転写、翻訳、細胞内局在、細胞外分泌の様式を検討した。CEA遺伝子ファミリーのクローニングを行ない、そのいくつかの構造を明らかにした。又CEAが細胞接着作用のある事を明らかにした。
6.膵癌、大腸癌に対する血清診断価値のあるモノクロナル抗体を作製した。また抗イディオタイプ抗体を作製し、診断、制癌への応用を検討した。
