| Research Abstract |
ヒトcーmyc遺伝子の2Kb上流に存在する自律増殖能を有するDNA配列(cーmycARS)を用いて試験管内DNA複製系を作製すべく研究を進めている。その過程でARSに特異的に結合するタンパク因子の精製を行った。ARS中のヘアピン構造を取り得る部位30ーmerを化学合成しセファロースに固定化してARSカラムを作製した。Rajc細胞核抽出物を硫安分画、ゲルろ過、モノQカラム(FPLC)で部分精製した後、ARSカラムにてARS特異的結合タンパクをアフィニティ精製したところ、分子量68Kのタンパクが精製された。このタンパク(p68)は0.3MNaCl存在下で特異的に上記30ーmerと結合し、他の塩基配列を有するDNAにより競合されなかった。p68はウェスタン・ブロッティングにより2種の抗ヒトcーmycモノクローナル抗体と反応する事からcーmycタンパクそのものである可能性が強く示唆された。抗cーmyc抗体はp68とDNAとの結合を強く抑制した。同抗体はcーmycARS複製系に加えると反応を抑制し、この阻害は抗原であるcーmycペプチドを添加する事により回復する事からcーmycタンパクは複製に深く関与している事が示唆された。p68とcーmycタンパクとの関連についてはペプチドマッピング及びタンパク一次構造の解析が進行中であり近い将来において明確となるであろう。p68がcーmycARSを用いた試験管内複製系においてどの様な役割を果しているのかに関しては、DNAポリメラーゼalpha・プライマーゼ、DNAヘリカーゼとの相互作用につき研究が進行中である。
