Research Abstract	膜蛋白質など疎水性蛋白質の結晶化法の確立を目指して、(1).蛋白質の構造安定性に及ぼす諸要因の把握、(2).結晶化条件の精密化,を柱に研究を行なった。研究対象としては、光合成関連蛋白質,チトクロ-ム酸化酵素,H^+ーATPase,バクテリオロドプシンならびにロドプシン・ファミリ-の諸蛋白質,べん毛モ-タ-,遺伝子組換え蛋白質RecA,それにアクチンを取り上げた。班活動の重点は,個々の蛋白質の結晶化において遭遇するであろう困難を議論し、互いに啓蒙し、励まし合うこと、とし、班員以外の方々の協力も得て研究会を開催した。日本における蛋白質結晶学はあまり盛んでないとばかり考えていたが、大盛況であった。考えを改めねばならない。今まで、潜在的な研究者の数を正確に把握していなかったのに過ぎない。ともかく、この研究分野は大いに発展するであろうと確信できた。この様な状況は、今まで孤軍奮闘の形で結晶化に取り組んできた研究者に多大な精神的援助を与えるものであり、この一点だけにおいても当班の存在価値はあったものと信ずる。また、班活動を通じて、新たな研究交流が生まれ,いくつかの共同研究が開始された。それらが実を結ぶまでにはかなりの時間を要するかも知れない。蛋白質の結晶化には地道な忍耐強い研究が必要であり,長期的な視野にたった研究支援体制を組織することが望まれる。個々の班員の研究成果については,班員間での研究の位相の差を考えると一律には評価しにくい。班全体で眺めるならば,結晶化に成功した膜蛋白質は4種類(バクテリオロドプシン,チトクロ-ム酸化酵素,光反応中心,F1ーATPaseのサブユニット)に増え、大きな前進であると考える。結晶化法に関しても、2種類の界面活性剤を組み合わせる方法や温度を変える方法が開発され、結晶安定化に働く添加剤が見いだされた。また、膜蛋白質の大量発現の成功など、価値ある研究も行なわれた。
