| Research Abstract |
1.PGE_2のアンタゴニスの中で、AH6809は、ラットではPGE_2后覚醒作用に培抗したがPGE_2の体温上昇作用には培抗しなかった。同物質は、サルにおいては、PGE_2の覚醒作用にも体温上昇作用にも培抗せず、別のPGE_2アンタゴニストが覚醒作用に選択的に培抗を示している。これらのことは、脳内のPGE_2レセプタ-の中に、サブタイプがあり、PGE_2の体温上昇作用と覚醒作用とに関与するサブタイプが異なること、これらのサブタイプに種差が存在することなどを示唆しているものと考えられる。
2.近赤外光を利用した田村システムを用いて、夜間睡眠中のサル前脳におけるヘモグロビンの酸化・還元状態を調ベたところ、レム睡眠時に特異的に、酸化型ヘモグロビンの増加と還元型ヘモグロビンの減少、総ヘモグロビン量の増加が認められた。レム睡眠中には、酸素の消費率が低下している可能性を示唆している。
3.無機セレン化合物はPGD合成酵素の活性を抑制する無機セレン化合物を脳室内に徴量投与すると、睡眠が顕著に抑制される。これは脳内内因性のPGD_2が主理的睡眠を引き起こしているという、われわれの仮説を強く支持する結果である。さらに、無機セレン化合物をラットに静注したところ、脳室内投与と同様の覚醒作用が得られた。投与量が多くなりすぎると、摂食量を抑制するなど副作用があるが、無機セレン化合物の過眠症に分類される諸種疾患への臨床応用は可能かもしれない。
4.冬眠リスの脳をサンプリングすることに成功し、冬眠期を含め、ほぼまる1年間にわたるリス脳のサンプリングが間なく完了しようというところである。各時期のリス脳のPGD_2、PGE_2、PGF_<2α>を定量し、PGの脳内での役割、冬期と睡眠との差異などについて考察する予定である。
