| Research Abstract |
タバコ葉緑体の遺伝情報系を構成する成分のうち核ゲノムにコ-ドされているものにつきその遺伝子をクロ-ン化して構造を明らかにするため、前年に引続いて解析を進めた。
1)50Sリボソ-ム蛋白のN末端アミノ酸配列を決定した結果、14KD蛋白のN末端がタバコ葉緑体DNAのORF55と一致し、これが大腸菌のL32と相同なため、rpl32と命名した。また,偽遺伝子と考えられていたrpl23の産物CL23をN末端より同定したのでこのrpl23が少なくともタバコでは真の遺伝子であることを明らかにした。
2)葉緑体の一本鎖DNA結合蛋白のうち、28KD,31KD,33KDの3種のcDNAを単離し、その一次構造を決定した。この結果いずれも2個のRNA結合ドメインを持っているので、これをリボヌクレオプロテインと分類した。さらに、無細胞系で3種の蛋白を合成し、核酸結合実験をしたところ、いずれもRNAと強く結合したことからこれらが真のリボヌクレオプロテインであることを実験的に証明した。
3)タバコ葉緑体より伸長因子EFーTuを精製し、このN末端アミノ酸配列を決定した。これよりcDNAを2種単離し一次構造を決定した。さらにゲノミック・クロ-ンを単離解析し、タバコには2種のETーTuの遺伝子が存在することを示した。両者はお互いによく似ているが、発現様式は異なっている。
