神経組織の加齢と内因性毒性物質(スフィンゴシン)の影響についての研究

| Research Abstract |
神経組織が加齢により変性する原因として我々は内因性毒性物質が関与している可能性を検討してきたが、本研究では中枢神経に多量に存在するスフィンゴ脂質の脂肪酸がないリゾ型スフィンゴ脂質(特にスフィンゴシン)に関して研究を行った。リゾ型スフィンゴ脂質は遊離アミノ基をもつことにより高い毒性を有し、数μMで細胞を死滅させる。我々はHPLCを用いてスフィンゴシンをはじめ、ガラクトシルスフィンゴシン、リゾスルファチドが正常脳内(ヒト、マウス)に存在することを明らかにした。このうち特にスフィンゴシンの濃度が高く、一般組織に100pmol/mg蛋白存在している。スフィンゴシンが非常に毒性が高く、その解毒機構を検討すると、スフィンゴシンは非常に速くアシル化され、セラミドになる。セラミドはもはや毒性をもたない。一方スフィンゴシンに糖鎖がつくと、ガラクトシルあるいはグルコシルスフィンゴシンになるが、これらの反応はマウスミクロゾ-ム内に存在するが、反応は遅く、Km値も高いことが明らかになった。
リゾ型スフィンゴ脂質のうち糖鎖の多量についたリゾガングリオシドについてはある種の疾患で増加していることが知られているが、我々はHPLCで超微量(数pmol)の測定法を開発した。正常ヒト、マウス脳内にも数pmolのリゾガングリオシドが存在していることが判った。スフィンゴシン、ガラクトシルスフィンゴシン、リゾスルファチド、リゾガングリオシドの加齢に伴う濃度の変化を現在検討中である。
