| Research Abstract |
It has long been believed that the major mechanism of action of antipsychotic drugs is based on the blockade of dopamine receptors in the treatment of schizophrenic patients. These pharmacological mechanisms has also been recognized as the adjustment of synergism between dopamineric and cholinergic neuronal activities. Recently, the mutual control of dopamine and methionine-enkephaline neurons has been shown. This study was aimed to elucidate further recognitions on the mechanisms of this action of antipsychotic drugs to the neuronal circuits of pre- and/or post-dopamine neurons, especially in the participation of enkephaline neuronal activity.
The proper or specific neuronal responses after acute administration of some agonists or antagonists were measured in the rat striatum and were suggested and estimated on the neuronal connections (circuits) around dopamine and enkephaline neurons from the present studies as following ;
1. GABA terminals ー> GABA-A receptors ー> Met-Enk neurons-> (DA, ACh, GABA) neurons
2. DA terminals -> DA2 receptors -> ACh neurons -> ACh/M1 receptors-> Met-Enk neurons -> (DA, 5HT, ACh, GABA) neurons
3. ACh terminals -> ACh/M2 receptors -> (DA, 5HT, ACh, GABA) NEURONS
4. DA terminals -> D2 receptors -> ACh neurons -> ACh/M2 receptors -> 5HT pre-synaptic terminals (projected from dorsal raphe nucleus)
5. GABA neurons -> GABA-A receptors -> pre-synaptic DA/D2 receptors -> ACh neurons
