| Research Abstract |
昨年の研究成果をふまえて研究を遂行し、次の成果を得た。
1.植物、土壊微生物、魚類、軟体動物について、ヒトと同様にLandsteiner現象が存在するか否か検討した。植物ではClerodendrom trichctomusの葉、花および茎に、土壌微生物ではProkopoa nagaiensisに、魚類ではサケとマスの卵に、軟体動物ではHelix pomatiaのアルブミン腺由来の分秘液にそれぞれLandsteiner現象が存在することを初めて明らかにした。これらの知見は、生物の進化を分子生物学的に解明するうえで極めて重要である。
2.Le^aーtransferase,XIVーtransfucosidase,HーαーDーgalactosetransferase,HーαーNーAcetylーDーgalactoーsaminetransferaseの活性の定量法を確立した。
3.血液型物質の安定性の経時的変化を知る目的で、フンボルト大学法医学研究所のOtto Prokop教授の助力を得てRobert Koch研究所から入手した手紙の封筒(Robert Kochが約90年前に弟子のFlosch教授に送ったもの)に附着していた唾液を用いて、ABO式血液型およびそれと類似の化学構造をもつLewis式血液型を鑑定した。吸収試験で調べた結果、Le(a+,b-),Oの結果がえられた。したがって、唾液中に分泌された微量の血液型物質は極めて安定なものであることを明らかにすることが出来た。
4.悪性腫瘍のある組織切片を用い、血液型物質の型転換(合成および分解)をスライドグラス上で行う方法を確立した。この実験方法を用いることにより、悪性腫瘍患者における局所の血液型物質の型転換が何故おこるかを解明することが出来た。
5.微生物由来のgalactosyltransferaseを用いて、組織の血液型物質の型転換が可能なことを明らかにすることにより、感染局所における組織の血液型物質の型転換機構を解明した。
6.血液遺伝学領域で注目されている有核細胞のDNA型別を行い、その応用を検討した。
