Research Abstract

Anti LHCP II 26.5 kDa apoprotein antibody and protein A-gold were used to localize the apoprotein in darkーgrown resting cells of Euglena gracilis var. Bacillaris and Z strain exposed to 150 ft-c of light to induce normal chloroplast development. An antibody reaction appeared first in the Golgi apparatus (at about 12 hr of light exposure) and as the reaction in the Golgi increased, deposition was seen over the plastid thylakoids. By 48 hr of light exposure both Golgi and thylakoids showed a strong reaction. There was no reaction in Golgi or thylakoids, however, when preimmune serum was substituted for antiserum. By 72-96 hr of light exposure, reaction in the thylakoids was maximal but disappeared completely from the Golgi. The results suggest, in agreement with data from synchronous cells and light-intensity shift-up experiments, that the nascent LHCP-apoprotein passes through the Golgi and may be processed there, on the way to deposition in the thylakoids. An antibody reaction was detected in the compartmentalized osmiophilic body (COS) before it was seen in Golgi or thylakoids.
