Research Abstract

1) 5' and 3' termini of rice dwarf virus (RDV), rice black-streaked dwarf virus (RBSDV), and rice ragged stunt virus (RRSV) genome segments were labeled with 32P. Individual segments were separated by electrophoresis, partially digested in alkaline, and subjected to sequence analyses by two dimentional electrophoresis.
2) Terminal structure of all genome segments of RDV was found to be 5'GGU/CAAA-----GAU3'.
3) Terminal structure of all genome segments of RBSDV was found to be 5'AAGUUUUU----GUC3'.
4) Terminal structure of all genome segments of RRSV was found to be 5'GAUAAA----GUGC3'.
5) All the genome segments of RDV, genome segment 10 of RBSDV, and genome segment 9 of RRSV had the segment specific inverted repeat at the 5' and 3' terminal regions.
6) The three plant reoviruses, each representing the three subgroups, were found to have virus specific terminally conserved sequences. These sequences were probably the signals for transcription and replication of viral genome RNAs.
7) The fact that all three plant reoviruses, each representing the three subgroups, have the inverted repeat in the genome terminal regions suggest that these structure play a key role in replication and packaging of viral genome RNAs.
