| Research Abstract |
タンパク質の機能性は、固有のアミノ酸配列とそれに基づく高次構造により発現される。本年度は、水産甲殻類アカフジツボ体液に含まれるレクチンを糖結合活性を持つ機能素材として利用するために、レクチンのアミノ酸残基側鎖を選択的に化学修飾して修飾率と凝集活性の変化を調べた。
140K、64Kレクチンのトリプトファン、リジン、チロシン、ヒスチジン、アルギニンの各側鎖を、Nープロモスクシンイミド、フルオレスセインイソチオシアネ-ト、Nーアセチルイミダゾ-ル、ジエチルピロ炭酸、シクロヘキサンジオンで修飾後、過剰試薬をゲル濾過または透折で除去した。試薬の添加の他は同じ条件で処理したレクチンをコントロ-ルとし、ウナギ赤血球に対する凝集活性を調べた。修飾率は吸収スペクトルあるいはアミノ酸分析で求めた。140K、64Kレクチンとも、トリプトファン、ヒスチジン、アルギニンの修飾により活性を失い、レクチン活性の発現にこれらのアミノ酸側鎖が重要な働きを演じていることを示唆している。チロシンとアミノ基の修飾では凝集活性の低下はみられなかった。従って、レクチンの機能素材化に当たっては、これらの側鎖を介した応用が考えられる。
64Kレクチンサブユニットは、128位に1残基のメチオニンを持つ。そこでブロムシアン分解により、そのペプチド結合を選択的に切断し、ゲル濾過クロマトグラフィ-で2つのジスルフイド結合で囲まれた糖認識結合ドメインを単離した。さらに、これを塩酸処理したセファロ-ス4Bカラムにかけてガラクト-ス結合性を保持したドメインを得た。
