新しい短期記憶実験法を用いての実験的健忘ラットに関する行動薬理学的研究

Research Abstract	本研究では遅延見本合わせ課題を用いてのmaze学習に焦点を当てた3ーpanel runway課題とoperant学習に焦点を当てた3ーlever operant課題を新たに考案し、ラットの短期記憶を客観的に測定する方法を確立した。3ーpanel runway課題を用いての実験では、scopolamineおよびAF64Aによりerror回数およびlatencyは有意に増加し、学習記憶障害が認められた。一方、3ーlever operant課題を用いての実験では、scopolamineは見本レバ-提示のtraining trialでの餌獲得には影響を及ぼさなかったが、3つのレバ-提示のあるtest trialでは著しく餌獲得を減少させ、短期記憶を障害した。嗅球摘出および背側海馬破壊によっても両学習課題は障害されたが、乳頭体破壊ではrunway課題でのみ障害された。scopolamineやischemiaによるこの学習・記憶障害はcholinesterase阻害剤tetrahydroaminoacridine(THA)やamiridin、コリン作動性薬物WEB 1881 FUおよびminaprineまたmethyl donorであるSーadenosylーLーmethionineによって改善された。以上の結果から、これらの障害はコリン神経系の機能抑制に基因していることが示唆される。薬物弁別法を用いての実験では、学習・記憶障害に対する改善作用を示したamiridin(10mg/kg)およびTHA(1.8mg/kg)の薬物弁別特性の解明を試みた。physostigmineおよびarecolineはTHAおよびamiridin弁別刺激に般化し、またTHAおよびamiridin間においても交叉的般化が認められた。dopamine agonist lisurideやamantadineは両弁別刺激に般化または部分的般化した。一方、両弁別刺激はともに抗コリン剤scopolamineによって拮抗されたが、haloperidol,SCH 23390およびsulpirideによっては拮抗されなかった。以上の結果から、THAおよびamiridin弁別刺激はともにACh神経系の機能亢進が最も重要な要因であるが、一部DA神経系の機能亢進も関与していることが示唆された。
