Research Abstract

IL1は免疫系のみならず、神経・内分泌系あるいは結合組織系に対しても増殖分化あるいは種々の炎症性メディエ-タ-の放出など多彩な作用を示す。ILー1リセプタ-はすでにそのcDNAのクロ-ニングやILー1リセプタ-桔抗物質の同定などもなされたが、ILー1を介するシグナル伝達の機序は依然不明の点が多い。この点に関し、(1)我々は従来からヒト白血病細胞株HSB.2サブクロ-ンを用いてILー1、ILー2およびIFNーγ産生における作用発現機序の解析を行っており、この成果を発表した(M&I、1989;Immunol.1990)。(2)さらに、ヒトアストロサイト細胞株(U373)がILー1に反応してILー6、ILー8およびMφの遊走と活性化因子(MCAF)遺伝子の発現が著明に誘導されることを見い出した。U373細胞におけるILー1のILー6mRNA発現誘導の機序にcAMPは直接関与していないが、ILー1の作用に対し調節的な役割をもつことを報告した(B.B.R.C.1990)。(3)ILー1によるU373細胞からのILー8、MCAFmRNA誘導は転写レベルでの制禦とmRNAの安定性の増強による知見を得ている(論文投稿中)。さらに、ILー1のELISA法による測定系を確立し、妊婦羊水及び慢性リウマチ患者血清と滑液中での測定を行ない、その病態意義について考察した(M&I,1990他)。(4)なお、サイトカインネットワ-クにおける他のサイトカインとの連関の重要性に鑑み、ILー2、ILー4、ILー5の作用機序の解析も並行して行った。とくに寄生虫感染やILー2投与におけるin vivoでの好酸球増多現象にはILー5が重要な役割をもっていることをILー5mRNA発現誘導あるいは特異的抗体による抑制などの実験によって明らかにした(J.Immunol.1990;Exp.Hematol.1990)。以上の研究成果はすでに関連の学会誌などに公表あるいは投稿中である。これらの研究はin vitroおよびin vivoにおけるサイトカインの作用と機能を一層明確にするにとどまらず、がん・感染・炎症における病態の免疫学的解釈の進展に貢献できたと確信する。
