| Research Abstract |
1.我々はこれまでに血清中リボヌクレア-ゼ(RNase)に遺伝的多型のあることを明らかにした。本年度は、遺伝的多型を示すRNaseの起源や分布を知るために各種臓器からRNaseを精製し、その性状を比較検討した。
(1)アセトン処理および一連のカラムクロマトグラフィ-を利用し、ヒト赤血球より2種類のRNaseを精製し、それぞれRNaseHE-1、HE-2と命名した。このうち含量の多いRNaseHE-1についてアミノ酸組成やN末端アミノ酸配列などのタンパク質化学的性状および酵素学的性状を調査したところ、RNaseHE-1は尿のnonsecretory RNaseと極めて類似していることが知られた。さらに、RNaseHE-1は、免疫学的にも尿nonsecretory RNaseと同一のものと考えられた。
(2)硫安分画および一連のカラムクロマトグラフィ-を利用し、ヒト脾臓より1種類のRNaseを精製し、RNaseHSと命名した。RNaseHSについて、アミノ酸組成やN末端アミノ酸配列などのタンパク質化学的性状および酵素学的性状を調べたところ、尿nonsecretory RNaseと極めて類似していた。また、免疫学的にも両者を区別し得なかった。しかし、RNaseHSと尿nonsecretory RNaseの糖組成間には明らかな相違が見られ、従って尿中RNaseの起源が脾臓ではないものと考えられた。
2.ヒト体液中に存在するデオキシリボヌクレア-ゼI(DNaseI)について抗DNase特異抗体を用いた板免疫ブロット法あるいは新たに開発した活性染色法と等電点電気泳動法を併用することによって、尿中DNaseIアイソザイムを再現性良く分離・検出することに成功した。日本人約400人の尿DNaseIを解析したところ、尿DNaseIは少なくとも10種類の表現型に分類することが出来た。詳細な人類遺伝的解析から、尿DNaseIは共優性な少なくとも4個の対立遺伝子によって決定されていることを明らかにした。
