| Research Abstract |
昨年度は、CCKーreceptor AntagonistであるCR1505がCCKー8による膵栄養効果を抑制することを見い出した。当該年度は このCRー1505がBHPによるハムスタ-実験膵癌の発癌を抑制し得るか否かを検索した。〈方法〉生後8週令の雄性syrian golden hamsterを用い、以下の4群を作成した。A・コントロ-ル群、B・CR1505投与群、C・BHP投与群、D・BHP+CR1505投与群。C群とD群にはBHP(500mg/kg体重)を週一回皮下注射した。また、B群とD群には、CR1505(100mg/kg体重)を週一回皮下注射した。CR1505の投与は9週まで行い、それ以後は対照液(生食)に変更した。各群を0〜9週、10〜14週、15〜19週、20〜24週と経時的に屠殺し、膵臓を摘出し、10%ホルマリンにて固定後、hematoxylinーeosin染色を行い光顕的に検索を行った。発生した病巣はhyperplasia、adenoma、carcinomaに分類した。有意差はX^2検定を用い、P<0.05を有意とした。〈成績〕 0〜9週では、各群においてcarcinomaの発生を認めなかったが、C群ではhyperplasiaを呈すものが87,5%存在したのに対し、D群では20%と有意に低かった。10〜14週ではcarcinomaの発生をC群で21,1%に認め、D群では22,1%であり有意差はなかった。しかし、adenomaの発生はC群で47,4%に認めたのに対し、D群では16,7%と有意に少なかった。15〜19週ではcarcinomaの発生をC群で47,1%に認め、D群では53,3%に認め有意差はなかった。20〜24週ではcarcinomaの発生をC群で80%に認めたのに対し、D群では50%であった。〈結論〉CCK receptor antaginstであるCR1505の投与にてBHPハムスタ-実験膵癌系において、前癌病変と考えられるhyperplasiaやadencmaの発生が有意抑制された。膵癌の発生に消化管ホルモンであるCCKが関与している可能性が相定された。
