卵胞の発育・分化に関与する新しい卵胞内因子の同定とその生理作用に関する基礎的研究

| Research Abstract |
我々は、1988年にブタ卵胞液中にインタ-ロイキン-2レセプタ-/p55(IL-2R/p55)誘導因子の存在を発見し報告したが(第18回日本免疫学会)、今回の研究により明らかにした点を以下に要約する。1.本因子が卵胞液中だけでなく、ブタ卵巣顆粒膜細胞の無血清培養上清中にも存在することを証明し、卵巣顆粒膜細胞で産生されることを明らかにした。2.卵胞液中のIL-2R/p55誘導活性は、卵胞の大きさだけでなく、ステロイド産生の観点からみた卵胞の発育とともに低下し、顆粒膜細胞からの産生も、卵胞の発育とともに低下することから、本因子は卵胞の発育に密接に関与する可能性を示した。3.卵胞液中のテストステロン/エストラジオ-ル比とIL-2R/p55誘導活性の間に有意な正の相関関係が認められ、本因子が卵胞の閉鎖に関連している可能性を示唆した。4.ブタ卵巣顆粒膜細胞の無血清培養上清中を限外濾過の後、ゲル濾過を行うことにより、本因子が分子量25,000から43,000の高分子で、しかも熱処理およびトリプシン処理により失活することから、蛋白成分に本活性が存在することを示唆した。5.ヒト卵胞液中にもIL-2R/p55誘導活性が存在し、さらに、本活性が抗インタ-ロイキン-1αまたはβ抗体によって中和されないことから、本因子が、すでにIL-2R/p55誘導活性を有することが判明しているインタ-ロイキン-1ではな、全く未知の因子である可能性を示唆した。6.卵胞液中に高濃度に存在するエストラジオ-ル、プロゲストロン、テストステロン、アンドロステンジオンにはIL-2R/p55誘導活性が認められないことを明らかにした。
以上の結果から本因子は卵巣の局所因子として、卵胞の分化・発育および局所防御機構に関与している可能性が示唆された。
