| Research Abstract |
1-methy1-4-phenyl-1,2,3,6,-tetrahydro pyridine(MPTP)をマウスに投与して作製したパ-キンソン病動物モデルを用い、大脳基底核における神経伝達物質の代謝と機能の変化を検討した。
1.MPTP投与に伴ない、黒質一線条体系ド-パミン作働性ニュ-ロンの破壊とド-パミン含量の急激な低下が認められたが、同時に線条体一黒質系GABA作働性ニュ-ロンの機能低下が認められた。
2.上記1のMPTP投与に伴なう黒質及び線条体におけるGABA含量の低下は、時間的経過、投与量依存性のいずれかの面から考えても、黒質-線条体系ド-パミン作働性ニュ-ロンの破壊に随伴する二次的・機能的な変化であると考えられる結果を得た。
3.MPTP投与に伴う線条体-黒質系GABAニュ-ロンの機能変化、特にGABA含量の低下の原因を神経化学的に追及したところ、GABA生合成の律速酵素(l-glutamate decarboxylase)やGABA分解酵素(GABA-Transaminase)の活性変化によるものではなく、GABAの代謝回転の低下に起因し、またこの変化のために、黒質内に存在する後シナプス性のGABA_Aレセプタ-複合体に過感作状態(up-regulation)が起ることが判った。
4.上記の各結果を基盤として、パ-キンソン病様症状を示す動物に対して抗パ-キンソン病薬としてのl-ド-パ、カルビド-パ、アマンタジンなどと共に、GABA_A受容体作働薬(ムシモ-ル、THIP、イソグバシンなど)を投与したところ、抗パ-キンソン病薬の作用を増強する傾向が認められた。従って、これらの増強効果発現機序について、更に詳しい解析を計画中である。
