Research Abstract

本研究班は、「大腸菌ゲノムの全体像」の中で「ゲノムの機能的全体像」を分担している。すなわち、大腸菌の遺伝子群の機能とその発現制御系のネットワークを拡大していって、ゲノム全体の機能を明らかにすることが最終の目標である。その手段として、従来の遺伝学的方法と、遺伝子の塩基配列の決定からの逆遺伝学的方法との二方向からのアプローチを進めている。本年度の研究成果は、遺伝子機能に関しては、ペリプラズム中の結合蛋白質依存性能動輪送系遺伝子群の塩基配列の決定から、リン酸レギュロンに属するsnーグリセロール3ーリン酸の能動輸送系が4種の蛋白質で構成され、結合蛋白質依存性能動輸送系に共通の構成であること(中田)、細胞の成長と分裂に関与するmra領域の遺伝子群と、その調節に関与するmra領域の遺伝子群と、その調節に関与するmre領域の遺伝子群の塩基配列を決定し、FtsW,MreC,MreDその他の蛋白質の生化学的役割を解析し(松橋)、熱ショックで発現するプロモーターを検索して、シグマ^<32>(rpoH)に依存して発現し、in vitroでシグマ^<32>を含む転写酵素によって転写されるプロモーターを6個同定し(由良)、また、ヒストン様蛋白質であるHU蛋白質は遺伝子組換えにも関与していること(加納)、H^+ATPaseのアルファ・サブユニットを多量生産させると大腸菌の分裂が異常になること(二井)などを明らかにした。また、遺伝子発現制御の分子機構に関しては、リン酸レギュロンの調節蛋白質であるPhoRがヒスチジン蛋白質キナーゼで、PhoB蛋白質をリン酸化し、リン酸化PhoBがリン酸レギュロンの転写を促進する、これと同様の機構をもつ制御系は現在まで(大腸菌では)約10組知られていること(牧野)、べん毛形成ではFliA蛋白質がシグマ因子活性をもち、べん毛レギュロンの後期オペロンの転写を促進し、PflB蛋白質がそのシグマ活性を阻害すること(沓掛)、cxo(チトクロームオキシダーゼ)オペロンが溶存酸素とcAMPによって調節され、cxoプロモーター領域に特異的な保存配列が存在すること(安楽)などを明らかにした。
