Research Abstract	1.分子模型の電子密度図への適合プログラムとしてはTOMを採用し、パ-ソナルIRISへの移植作業は完了した。2.大型計算機上で、蛋白質結晶の構造の最小2乗法精密化プログラム(KonnertーHendricksonのプログラムを改良したもの)の実行速度をできるだけ速める徹底的な最適化を行い、それをパ-ソナルIRISに移し、その移植を完了し、上のTOMと有機的に結びつけて使用できるシステムを構築した。3.リボヌクレア-ゼMs3'ーGMP複合体結晶の分子置換法による解析に関しては、前年度に決定することができたタンパク質分子の格子軸に対するオリエンテ-ションおよび位置から導かれたあらいスタ-ティング・モデルから出発して、これまで構築してきた(TOM+精密化)システムを用いて精密化を行った。作り上げたシステムは、まさしく効率的に働き、大型計算機をベ-スとした従来のやり方の少なくとも5倍は能率よく精密化を行うことができるようになった。それを示すものとして、我々はシステム構築後たった1年で(1)RNaseMs3'ーGMP複合体(2)サブチリシンーSSI複合体(3)RNase Ms2'ーGMP複合体(4)RNase MsGfpC複合体(5)トリプシンーSSI複合体(6)インタ-フェロンβ(7)RNase Rhの7種もの多数の結晶に対する精密化に成功したのである。従来は、精密化に半年〜数年かかっていたのが普通であり、大幅に短縮され訳である。今後、使用経験をより増すことによって、システムをより使いやすいものに作り上げていく方針である。
