Research Abstract	初年度は癌細胞における細胞増殖の脱制御の機構をDNA複製の観点より解明するための実験系の開発に力を注ぎ次の様な成果を得た。 a)試験管内DNA複製系:現在最も進んだモデル系であるSV40DNA複製系をさらに一歩進めてヌクレオソ-ム構造を有するSV40DNAミニクロモソ-ム複製系を、DNAポリメラ-ゼα及びδを用いて構築した。この系を用いて複製に際しヌクレオソ-ムは2本の娘鎖DNAに均等に配分される事を明らかにした。この系で高次の複製調節の解明が期待される(花岡)。一方ヒトDNA複製開始領域の有力候補であるcーmycARSを用いて作製したトランスジェニックマウスではcーmycARSは全組織に分布し、再回収したプラスミドに512bpのマウス由来配列が附加し、これが複製を強く促進する複製エンハンサ-である事を明らかにした(有賀)。cーmycARSのcoreと考えられる配列に結合するタンパクをRaji細胞抽出物からアフィニティカラムにより精製し分子量68kタンパク(p65)を得た。p68のDNAに対する特異的結合はcーmyc抗体により阻害され、又p68は同抗体と反応する事からcーmycタンパクである事が強く示唆された(吉田)。b)調節因子:ヒト肝癌に密接に係わるHBV・Xタンパクはcーmyc遺伝子のトランス活性化因子として作用する事が見出されその機序に興味が持たれる(小池)。他方、癌抑制遺伝子RBを昆虫細胞で過剰発現させる事に成功し、RBタンパクと親和性の高い2種の細胞性タンパクを同定した(秋山)。増殖抑制との観点からこれらの機能解明が待たれるが、遺伝生化学的に増殖抑制を解明する系として、酵母接合因子が酵母膜受容体、細胞内情報伝達系を経てCDC28/CDC2キナ-ゼによる増殖サイクルの制御、さらにCDC7キナ-ゼによる複製複合体に対する直接制御を解明する事が各遺伝子を単離する事により可能となった(新井)。
