Research Abstract

今年度得た、新しい知見と成果は、次の通りである。1.精子の運動開始に、海産魚ではCa^<2+>、pHが重要な役割を果し、サケ科魚類では、特異的プロテア-ゼが関与する。サケ科魚類類精子の運動能獲得は、運動装置である鞭毛軸糸のみならず、細胞膜での変化を伴う。(森沢)2.哺乳類精子の受精能獲得は、頚管粘液で促進される。先体反応は、透明帯で誘起されるが卵丘は関与しない。(佐藤)3.ハムスタ-卵管より精製したオビダクチンは、卵胞卵の受精率を著しく高めた。(内貴)4.ブタおよびウシの卵胞液中には、卵の雄性前核形成能を高める高分子成分が存在する。(豊田)5.ヒトデ卵ゼリ-より精製した先体反応誘起物質は、新規糖鎖を含み、精子に特異的に結合する。(星)6.ブタ透明帯糖タンパク質のAsn結合型中性糖鎖の構造を明らかにした。(高崎)7.マウス精子の頭部細胞質に、プロテインキナ-ゼCアイシザイムのII型が発現している。(鈴木)8.ヒトおよびマウスで、アクロシン遺伝子を解析し、アクロシンの一次構造を決定した。この遺伝子には、雄性生殖細胞特異的に発現する遺伝子に含まれているGGGTGGG配列が認められた。転写、翻訳は、精母細胞パキテン期にはすでに始っていた。(新井)9.精子核の塩基性タンパクの遺伝子解析を、ヒキガエル(プロタミン)、ツメガエル(SP4)につき行い、両タンパクの一次構造を決定した。両者とも精子形成の最終段階で合成されるが、遺伝子の転写は、前者では精細胞、後者では精母細胞で行われる。(片桐)10.カエルの精子前核形成における精子核塩基性タンパクの離脱は、卵細胞質中のヌクレオプラスミンによって非酵素的に行われる。(片桐)11.ウシ輸卵管上皮細胞を腺構造を維持したままで一月以上培養する方法(内貴)、アフリカツメガエル体腔卵を受精させる方法(片桐)、Y染色体部分欠失マウス精子を効率良く受精させる方法(豊田)をそれぞれ開発した。
