Research Abstract	本年度は、Ca^<2+>シグナル系及びイノシト-ル燐脂質(PI)代謝に関与する種々の蛋白質の生化学的及び分子生物学的研究に加えて、これらの蛋白質が関与する種々の現象に関する薬理学的及び生理学的研究が行われた。 (1)低分子量G蛋白質ーホスホリパ-ゼ(PLC)系:ボツリヌス毒素が低分子量G蛋白質をADPーリボシル化するとともにリンパ球PLCの活性化を誘導することを発見し(大沢)、PLCがATP依存性に活性化されること、この活性化がイソフラボン化合物で抑制される事を見いだした(小河原)。PLCγ_2のZ領域がチロシンキナ-ゼやリン酸化蛋白の認識と活性化を行う部位であることを見いだした(竹縄)。(2)イノシト-ルリン酸認識蛋白質:IP_3、IP_4の分解に関与するフォスファタ-ゼに分子多様性を見いだした(中川)。IP_4アナログを化学合成し、IP_4認識蛋白質の活性中心の構造解析を行った(古賀)。(3)プロテインキナ-ゼC(PKC)アイソザイムとその局在:免疫電顕法を用いてPKC各分子種の神経細胞内特異分布を明らかにした。PKC各分子種に加え、カルモジュリン依存性酵素であるカルシニュ-リン(Aα、Aβ)のmRNAの脳内分布をin situ hybridization法により明らかにした(田中)。PKCアイソザイム間の機能的差異を、PKC過剰発現細胞を用いて転写活性化能を指標とする事により明らかにした(大野)。PKC分解産物に分子量8万の新しい活性化型酵素の存在を明らかにした(山村)。PKC分子種の活性変動を蛋白、mRNAレベルで解析し、それぞれが異なる細胞周期で機能する事を示唆し得た(荻田)。 (4)Ca^<2+>結合蛋白質及びフォスファタ-ゼの特性:合成カルモジュリン阻害剤をリガンドとしたアフィニティ-精製法により新しいEFーhand蛋白質を見いだした(日高)。プロテインフォスファタ-ゼ2AのδサブユニットがPKCにより燐酸化されることを見いだした(武田)。(5)PI代謝ーPKCの機能:単離前庭神経核細胞を用いた実験により、PKCのチャンネル制御機構における役割を示唆した(笹)。PI代謝に共役したグルタミン酸受容体が、海馬CA3シナプスにおける長期増強において重要な役割をはたしていることを示唆する結果を得た(杉山)。Balb/c3T3細胞においてインスリン様成長因子(IGFーI)により細胞内Ca^<2+>濃度の上昇及びPKCの持続的活性化が惹起されることを見いだした(尾形)。グリコフォリンー4、1蛋白質を介する赤血球膜変形能の調節にPI代謝が関与していることを明らかにした(石橋)。これら研究班員の研究成果は国内外の学術誌に掲載され、平成3年1月10〜11日に東京で開催された公開シンポジウム及び合同班会議で公表され、情報交換を行った。
