Research Abstract

最近シンクロトロン放射光の強力X線を利用した回折・散乱実験が急速に進展し、結晶中での酵素反応を高時間分解能で解析する研究が始まり、また筋収縮の分子的機構や膜蛋白機能の解明への動的構造解析の貢献も大きくなりつヽある。本総合研究はシンクロトロン放射光の利用者とこれに強い関心を持つ研究者を中心に種々の蛋白質や生体中の超分子構造の動的構造解析について現在の問題点を明らかにし、日本のこの分野の研究を促進させることを目的とする。
本年度はまず放射光X線回折系を高精度で利用するべく、CCDを用いたX線回折用2次元検出器、蛋白質結晶解析全自動回折計の開発等に着手した。結晶解析については3ーイソプロピルリンゴ酸脱水素酵素のキメラ分子及びダンシルリジン結合免疫グロブリンFv鎖を結晶化し、構造解析を行なっている。また、ATP合成酵素の動的構造解析、モザイクウイルス蛋白質の会合の速度定数の決定を行なった。特記事項としてATP単結晶が結晶状態のまヽ二水物と三水物の可逆変化し、隣接分子の構造が変化をすることが見つかった。筋肉については、まず、ミオシン頭部の構造がATP等により変化し、また無負荷短縮中の筋肉でも相当数のミオシン頭部がアクチンフィラメント周辺にあることが明らかになった。またミオシン頭部のPDM化学修飾により赤道反射強度の減少と143A^^°子午線反射の増大が観測された。更に筋肉に正弦波的長さの変化を与えたときのミオシン頭部の運動をモデル計算で説明できた。生体膜関係では、まずリン脂質膜で出現するリップ構造の周期の温度依存性を明らかにし、その理論的考察を行なった。また生きたホタルイカ視細胞から得たX線回折像をモデル計算およびパタ-ソン関数で解析した。紫膜では光プロトンポンプ作用に重要なM中間体の構造を明らかにした。大小規模の研究打ち合せ会を6回行い、以上の研究を綿密にかつ総合的に進めた。
