蛋白質チロシン残基の燐酸化とその細胞生物学的意義

Research Abstract

1)src familyチロシンキナーゼに関して、pp60^<c-src>を燐酸化するc-src kinase(CSK)の細胞機能における役割を明かにするため、CSK遺伝子を欠損させた動物の作成したところ、CSK欠損個体は胎生11日目で到死となった。その胚を検索すると神経管の発達不全、頭部神経冠細胞の細胞死が確認された。またこれらの組織においてpp60^<c-src>、pp59^<fyn>、po53/56^<lyn>の顕著な活性化が認められた。これらの結果はsrc familyは神経系の発生過程に何らかの重要な働きをしていることを示唆している。またp60^<c-yes>、pp59^<fyn>の自己燐酸化による活性化について検討し分子間反応であることを明かにした。さらに正常細胞における他のsrc familyチロシンキナーゼに関してfynやlynの標的蛋白質について検討した結果、lynの下流にはp75^<HS1>が存在すること、fynの下流にはVavやPLC-γ、MAPキナーゼ等の存在を示唆することが出来た。新しく見いだされた非受容体型チロシンキナーゼであるp72sykは血小板においてpp60^<c-src>の約半量存在し、生理的なリガンドであるスロンビンによって活性化されるがその活性化解除においてはCa^<2+>の存在が必要である。このCa^<2+>の作用は阻害剤の実験からミオシン軽鎖キナーゼが関与している可能性が強い。2)増殖因子受容体チロシンキナーゼにおいてはインスリンで細胞を刺激するとインスリン受容体キナーゼ依存性にイノシトール燐脂質3キナーゼの85Kサブユニットのチロシン残基が燐酸化されることを明かにした。さらにその燐酸化部位は368,580,607番目のチロシン残基である。またインスリン受容体キナーゼはIRS(Insulin-receptor substrate-1)を燐酸化しその燐酸化部位をイノシトール燐脂質3キナーゼの85KのSH2の領域が認識する可能性を示唆した。さらに上皮成長因子受容体チロシンキナーゼの下流にG蛋白質に関係するGAP(GTPase activating protein)が存在し、この系が細胞の増殖分化において何らかの重要な役割を演じていることを示唆した。3)チロシンホスファターゼに関して肝臓において4種類(L1-4)分離同定することが出来た。そのうちL1は2つのSH2領域をもつ全く新しいホスファターゼであることを明かにした。
