Research Abstract

本研究組織はヒト主要組織適合抗原複合体(HLA)の構造、発現調節、生物学的機能および臓器移植における拒絶反応の機序を解析することにより集団遺伝学、進化遺伝学、免疫遺伝学、臨床遺伝学および臓器移植学の分野でHLAを総合的に研究するために編成され、補助金は以上の目的を達するための研究費ならびに会議費として使用された。本年度の成果として以下の項目が上げられる。
(1)HLAの研究材料として必須であるB細胞株、単クロ-ン抗体、L細胞トランスフェクタント等が、全世界より収集され、これを必要とする研究者に、供給するシステムが整備された。(2)HLA領域内に新たな遺伝子の存在が確認され、これを同定した。(3)HLA分子の多型を電気泳動法で同定するシステムを確立した。(4)HLAクラスII対立遺伝子をDNAレベルで決定する方法が確立され、日本人集団に存在する対立遺伝子のほぼすべてを同定した。(5)HLA分子を認識するT細胞レセプタ-を、DNAレベルで同定する方法を開発し解析した。(6)HLA遺伝子を細胞やマウス個体に導入し、機能を有する形で発現させることに成功した。また、これらを応用して、ウィルス抗原あるいは細菌抗原に対する免疫応答における、HLA分子の機能を解析し成果を納めた。(7)各種自己免疫疾患患者のHLAを、DNAレベルで決定し、HLA領域に存在し疾病感受性に影響を及ぼす遺伝子を同定した。(8)臓器移植の生着率に及ぼす、HLAーDP対立遺伝子あるいはマイナ-組織適合抗原の不一致の影響を解析した。
