Research Abstract

構造解析:PCR法で増幅したHLAクラスII遺伝子を解析することにより多型をDNAレベルで同定した。HLA分子の蛋白レベルでの多型を、一次元等電点電気泳動により解析し、14種類の新しいクラスI分子を同定した。60kDaの単球に特異的なアロ抗原を識別する単クロ-ン抗体を作製し、これをコ-ドする遺伝子がHLAと連鎖していることを明らかにした。免疫グロブリン超遺伝子族に属する遺伝子の分子進化を解析するプログラムを作製した。機能解析:抗原ペプチドのHLAクラスII分子への結合性に関する多型を明らかにし、T細胞性免疫応答との相関を証明した。HLAーB5あるいはHLAーDR分子上の一個のアミノ酸置換を識別するアロ特異的T細胞のTレセプタ-が予想外に多様であることを明らかにした。自己の抗原提示細胞上に発現されたHLAーB分子を特異的に認識して、これを傷害することにより抗原特異的CD4^+T細胞の増殖を抑制する、自己細胞傷害性CD8^+T細胞の存在を明らかにした。さらにHLAーDRαトランスジェニックマウスでDRαEβ分子の発現の組織特異性が異なる3系統のマウスを樹立し、胸腺実質細胞でのみMHCクラスII分子が発現されている場合にも、自己MHC反応性T細胞レパトアが欠失し得ることを発見した。疾病及び移植とHLA:原田氏病、インスリン自己免疫症およびインスリン依存型糖尿病と強く相関するHLAクラスII遺伝子をDNAレベルで同定した。また腎移植においてHLAクラスII遺伝子のDNAタイピングの一致が生着率の向上に重要であることを明らかにした。
以上のように、HLAに関して総合的研究がなされ、特に分子遺伝学的アプロ-チの導入により、HLA多型の構造、起源および機能に関して多くの成果を納めることが出来た。
