| Research Abstract |
バフンウニ精巣よりチオシアン酸グアニジン法により全RNAを抽出しオリゴ(dT)セルロ-スを用いてpoly(A)^+RNAを調製し,cDNAを合成,ファ-ジベクタ-としてλgt10およびλgt11を用いてcDNAライブラリ-を作製した。λgt10を用いて作製したライブラリ-は独立クロ-ン60万,λgt11を用いて作製したライブラリ-は独立クロ-ン200万のもので1kb・p〜7kb・pのインサ-トを含むよいものであった。このライブラリ-を用いて(1)膜結合グアニレ-トシクラ-ゼ,(2)精子活性仕ペプチドI,(3)cGMPホスホジエスラ-ゼ,(4)WGAレクチン結合タンパク質に対するオリゴヌクレオチドプロ-ブを合成し,cDNAのスクリ-ニングを行なった。(1)〜(3)のタンパク質に関してはホモロジ-検索により,(4)のタンパク質については精製したタンパク質のプロテア-ゼ分解によって得られたペプチド断片のアミノ酸配列からプロ-ブを作製した。(1)のタンパク質についてはfull lengthのcDNAも単離,(2)については約50のクロ-ンを(3)については20のクロ-ンを,(4)については50のクロ-ンを得ており,現在cDNAの塩基配列の決定を進めている。
バフンウニ精子およびタコノマクラ精子について,それぞれ特異的に作用する精子活性化ペプチドをヨ-ド化し,結合実験,架橋実験を行ないそれぞれ,少なくとも2種類の受容体タンパク質が存在することを明らかにした。タコノマクラ精子については高親和性,低親和性受容体が存在し,高親和性受容体は精子の呼吸,運動性の促進に関与していること低親和性受容体はcGMPの産生を促進する情報伝達機構の活性化に関与していることを示唆する結果を得ている。
バフンウニ精子よりリン酸化型,脱リン酸化型グアニレ-トシクラ-ゼを精製した。
