| Research Abstract |
Lーグルタミン酸受容体は現在まで(1)イオンチャンネル型と(2)代謝調節型に大別され、さら前者はNMDA,カイニン酸,キスカル酸型のサブタイプに分類されている。先の合成した4種の2ーカルボキシシクロプロピルグリシン類(CCG)のうち、CCGーIVがNMDA型受容体の選択的かつ強力なアゴニストであることから、この受容体におけるグルタミン酸の活性座はfolded型であると結論された。平成2年度はこれらの結果をもとに、グルタミン酸のαーアミノ酸部位の空間的情報、受容体単離のためのリガンドの設計,三員環の空間的役割の検討などを目的とし、三員環上に置換基を導入した類縁体の合成を行った。
Dーセリンを共通の出発物質として用い、EあるいはZ型のアリルアルコ-ルの分子内シクロプロパン化を鍵段階としてfolded型のCCGに対応する4種のメトキシメチル体(cis及びtransーMCGーIII,IV)を立体選択的に合成した。母核としてしCCGーIVをもつMCGのうち、cーMCGーIVはαーアミノ酸部位の配座が固定されており、これがCCGーIVと同じNMDA型を活性化することから、この受容体におけるグルタミン酸の活性配座を図のように特定できた。一方、tーMCGーIVはNMDA型ではなくカイニン酸型を活性化し、この受容体もfoldedの立体配座を認識する事がわかった。
