Research Abstract

1)ヒト赤血球膜グルコ-ス輸送体分子を抗原として作製した抗体と、HepG2グルコ-ス輸送体C末の合成ペプチドに対する抗体の2種を用いて、GLUT1(赤血球/HepG2型グルコ-ス輸送体)の分布をイムノブロット法と凍結切片の免疫組織化学染色により検索した。
閉鎖帯によって結合された血管内皮細胞を有する血液一組織関門部(脳、網膜、虹彩、末梢神経)では内皮細胞がGLUT1陽性であった。陽性部位は細胞膜の全周にわたった。一方、血液一組織関門の存在しない組織の内皮細胞細胞膜はGLUT1陰性であった。閉鎖帯によって結合された上皮細胞が血液一組織関門を形成する場合(網膜色素上皮、毛様態、脈絡叢、抹消血管神経周膜)では、関門をなす上皮細胞細胞膜がGLUT1陽性であった。また胎盤関門を形成する合胞体性栄養膜の細胞膜もGLUT1陽性であった。このように、GLUT1グルコ-ス輸送体は血液一組織関門部に豊富に存在し、グルコ-スの関門通過を司っていると思われる。今後は、他の4つのisoforms(GLUT1ーGLUT5)の存在の有無についても検討を加えていきたい。
2)表皮の形態形成及び角質化の過程は、過剰量のビタミンA(V.A.,20μM)投与により抑制され、さらにin vitroでは表皮細胞の粘膜化生が誘導される。このV.A.によりさらに促進され、proteinーtyrosine kinase阻害剤(herbimycin A)により抑制されることを見いだし、これらの表皮を形態学的に検討した。
3)胎仔精巣の形成に伴いセルトリ細胞の基底側細胞質内にアクチンフィラメントが集積し、その後中間径フィラメントが発達する。未分化精巣の器官培養系を用い、サイトカラシンD、コルセミドの効果を光顕及び電顕的に観察し、セルトリ細胞の分化(上皮様の極性化)に伴う細胞内骨格系の役割を検討した。また、基底膜、アクチン及び中間径フィラメントに対するサイトカラシンDの影響を免疫組織化学的に調べた。
