重症筋無力症におけるアセチルコリン受容体γ/δサブユニット遺伝子領域の変異の解析

Research Abstract	1.アセチルコリン受容体γ/δサブユニット遺伝子とDNAポリメラ-ゼβ遺伝子に関する研究:アセチルコリン受容体γ/δサブユニット遺伝子はヒト第2染色体長腕のq32ーqterにマップされており、ほぼ同じ領域にマップされるγ/クリスタリン遺伝子クラスタ-の異常が自己免疫疾患の疑われる患者において見出された。当初、アセチルコリン受容体γ/δサブユニット遺伝子にも異常があると考えられたが、サザンブロット解析ではこの患者のアセチルコリン受容体γ/δサブユニット遺伝子に異常は見出されず、意外なことにDNAポリメラ-ゼβ遺伝子の異常が検出された。DNAポリメラ-ゼβ遺伝子は主に脳と胸腺で強く発現されており、T細胞レセプタ-の遺伝子組替え後の修復に関与していると考えられることから、この遺伝子異常はT細胞レセプタ-の異常として反映される可能性がある。 2.胸腺腫好発BUF/Mnaラットの分子遺伝学的解析:重症筋無力症における胸腺腫の公症の機序を解析する目的でヒト胸腺腫のモデル動物BUF/Mnaラットの分子遺伝学的解析を行なった。このBUF/Mnaラットの染色体は不安定であることが当られているので、セントロメアのサテライトIDNAに変異がないか否かをその塩基配列を決定して調べた。25クロ-ンの塩基配列を決定したところ変異クロ-ンを1つ発現した。このクロ-ンは動原体構成蛋白が結合すると推定されるTGGGAAという配列を持っておらず、動原体としての機能は持たないと考えられる。この変異クロ-ンは姉妹染色分体間で不等交差が生じた後、数回の相同染色体間のgene conversion(遺伝子変換)を経てコピ-数が増幅したもので、染色体の不安定因子となると考えられる。ヒト胸腺腫にも同様の変化がアルファサテライトDNAに存在するか否かは今後の興味ある研究課題である。
