| Research Abstract |
遺伝性脱髄疾患の一つにgalactosylceramidase Iが遺伝的に欠損しているKrabbe病がある。我々はすでにgalactosylceramideの分解は二つの酵素で触媒され、そのうちgalactosylceramidase IのみがGLDで欠損し、IIは正常であることを報告した。Galactosylceramidase Iは精製が困難で、未だ完全に精製されていない。我々はブタ賢臓よりgalactosylceramidase Iを約10000倍に精製し、SDSーPAGEにて分子量約40000と30000の主要バンドがあることが明らかにした。この蛋白質のどちらがgalactosylceramidase Iかは不明であるため、それぞれの蛋白質を切り出し、N末端のアミノ酸配列を40KDaの蛋白のみ決定したが30KDaの蛋白はN末端がブロックされていた。現在エンドペプチダ-ゼ処理の後、中間部のアミノ酸配列を決定中である。
一方、cDNAクロ-ニングのためブタ賢臓よりcDNAライブラリ-を作製し、N末端アミノ酸配列よりプロ-ブを作製し、スクリ-ニングを行っている最中である。
