血管内皮細胞におけるトロンボモジュリンの発現に関する研究

| Research Abstract |
培養臍帯静脈血管内皮細胞(HUVEC)とヒト巨核芽球性白血病細胞(MEGO1)を用い、次のことを明らかにした。
1)HUVEC、MEGO1ともに3〜5mMのdbcAMPに応反して、HUVECの場合は約250%、MEGO1の場合は約600%thrombomodulinが増加した。
2)その他の細胞内cAMPを増加させる薬剤IBMX、forskolin、PGI_2でも同様の効果が観察された。
3)ILー1、TNFαでは、thrombomodulinは逆に50%減少した。
4)thrombomodulin遺伝子の上流には、cAMP responsive elementは見いだし得なかった。
5)ヒトthrombomodulinのDNAをCHO細胞に組み替え、組み替え体のthrombomodulin(γーTM)を発現させてところ、いままで知られていた分子量サイズのthormbomodulin(MW:約750,000)の他に、高分子のthrombomodulin(MW:170,000)が発現した。ここで通常のものをtype1、高分子のものをtype2 TMと命名すると、組み替え体CHOの場合、type1/type2はほぼ1/1であった。
6)次にimmunoblot法で、HUVECにおけるtype1/type2の発現を調べた。通常の培養条件でも、HUVECにtype2が約5%発現していた。
7)HUVECを、cAMP、フォルボ-ルエステルで刺激すると約12時間後に、type2が約20%にまで増加した。
